Once you collect the sisters in an area the daddies will still come around. If you haven't collected the sister yet, sometimes you need to follow a big daddy to the hole in the wall where he will call the sister out, then you kill him.

In one level there's a glitch that says there is still a sister left in the level even though you have actualy gotten them all (actually there's an extra sister) and this led me to get real pissy looking for that damn sister that wasn't there.

It's in Neptune's Bounty, and I don't think the glitch has to do with the game saying there's an extra sister, but rather you being able to find a 4th sister when there are only 3 sister icons on the map screen. If you're lucky enough to find the extra sister, a 4th icon appears on the map screen(or pause screen, whichever has the sister icons.)

NOw I remember. There are three sister icons, but if the glitch is triggered you can find a fourth. If you do, when you try to leave the level ,it will tell you that all of the sisters in the level have not been found, because the game only recognizes if you have found the same number of sisters as there are icons. It sees you getting an extra sister the same as if you only found two.

oh yeah. Got my first sister rescue present. A fuckload of adam, a control big daddy plasmid and somthing else (I forget)

I like killing the rosies a lot better than the bouncer daddys. At least you can dodge what the rosies are dishing out. The bouncers are just insane.

I just got freeze ray too. Freezing enemies and smashing them is awesome! I also picked up the target dummy plasmid. Is it very useful?

New question, when are you supposed to get the research camera? I scoured the medical clinic and the first level but never received it.

It's canon during the story, you can't miss it. If you're fighting Rosies, you must be in Neptune's Bounty, so you shouldn't be too far at all from talking to the character who gives you the camera.

sweet. I was worried I missed it somehow.

I noticed another thing too. In the first big room on neptune's bounty, the second I come through the door, there's a rosie right there. He heads back into the hallway I came from and gets a sister out of the hole in the wall. I kill him, rescue the sister, etc...

Next I am (obviously) low on ammo and medkits, so I head back to the vending machine at the start of the level. When I return to the big room where I formerly killed the rosie, there's another rosie, with another sister. I kill the rosie and rescue the second sister...which makes 2 sisters total rescued at the exact same location on neptune's bounty level. My menu still shows 3 sister icons at this point. 2 rescued and 1 still to go.

Is this the glitch or is that normal? If I'm at a daddy spawn point (like that first big room of N's B ) and it is also near a hole in the wall, will respawned daddies just keep going to the same hole in the wall and haul out little sisters until there's none left on the level? Or should sisters be coming out at different locations on the level? For instance, could I just sit there and wait for new rosies to get new little sisters at the same location until all of the sisters for that level are exhausted?

Look at a few of the previous posts in this topic. RLY and myself elaborated a bit on this particular 'extra sister glitch'. Yes, you found an extra sister. IIRC, there are two places in N's B where the extra sister could spawn and where you found your second sister is one of them.

When it comes to the target dummy plasmid that you mentioned getting earlier, I can't say I ever bought it or used it. I always saved my adam for the sure things and my tactics throughout Bioshock focused on hacking. I always hacked each and everything turret and camera - which got me by plenty of dangerous areas since once I hacked everything, I could run back through the turret area leading splicers to their doom.

I second the hacking. Buy any plasmid that makes hacking easier, and hack the shit out of everything.

I've not been hacking the security systems very much...but that first big room on N's B is one place where I really wish I did. There are new splicer spawns on that room every time I go through and return via one of the exit doors. If the turrets there were under my control I can see how I would really benefit in that particular location.

Same on the medical pavilion main room. I suppose they help a small amount with the big daddies too eh?

I really have a hard time with any of the security systems harder than about 1/3 difficulty. The water just comes through the pipe too quickly. Any good tips for hacking?

Uncover as many of the hidden squares as you can before the water becomes a threat, then just go fast. Also stock up on any plasmids that enhance hacking. I heavily relied on plasmids that: healed health or eve each time I hacked something; reduced the speed of the water; reduced the number of alarms; etc. Also stock up on instant hacking tools when you can. Those are extremely useful for the random expert level safe in early areas.

ETA:

Other tips I, personally, approve of:
-ignore most armor piercing rounds you find or at least don't spend any money on them. Early game, electricity is always more powerful and late game, you have enough money to buy your fill of any ammo you want(of course, by this time you'll have the crossbow, which will make you scoff at most of your other weapons)

-keep your eyes heavily peeled for Power to the People kiosks.

-Once you find the Telekinesis plasmid, use it for fighting. Throwing corpses is almost always a 1 hit KO until you reach nastier enemies, and throwing most things is more powerful than some of your weapons(it also doesn't eat a ton of eve, like electroshock does). For example, I once beat a miniboss and his entire room of minions and turrets with a handcart I accidentally set on fire and just threw around with TK.

I can't emphasise enough on the Power to the People kiosks, if you don't look for them it makes late-game really hard if you are depending on weapons.
For the hacking part, I get the plasmid that gives me health every time I successfully hack something. Also something to note is that you can't die from hacking (on the vanilla version, I don't know if they've patched this), so just keep hacking something if you are on low health until it's successful. Get practice on everything; the algorithm for the random generation of puzzles is pretty stupid since after a while you really know what the path is. I had in problems in mid-late game hacking everything on my first go.

Electroshock is pretty useless unless there's water and a lot of enemy so you can headshot them all. I find it only useful to stun the turrents and hack them if they are far away. Other than that, it uses tonnes of Eve and eve gets pretty rare in Hard mode.

Telekinesis is the underrated dog of the game, you can use it to find all sorts of goodies in random places you'd never thought to look (roofs, cages etc) besides what Tarcanus said.

Once you get Adam try saving it before buying all in a go, and sometimes the plasmid selection they give you is stupid. For example I didn't get that plasmid that freezes people until lategame, but I got it for my Wrench so it freezes people when I thwack them, this way it doesn't use Eve.

for shooting the big daddies, does it help to aim for the head or are they equally tough to kill no matter where you aim.

I know most splicers seem to tolerate the heashots less than the body shots.

oh man, had a fun game last night.

I started hacking the bajesus out of everything and it is waaay more fun to have turrets on your side. I also got the security bullseye plasmid, which I've been using to some effect on the big daddies. Those splicers that jump up into the ceiling though....houdini splicers??

Tough bastards

got the research camera too.

I find I'm going through this game really really slowly...jumping at everything and retracing my steps a lot. Is that pretty normal?

The splicers that jump to the ceiling are Spider Splicers. You'll know the Houdinis when you see them, haha. Eventually, if you take enough pictures of spider splicers, you get a plasmid that allows you to use spider splicer organs as health kits, so you wind up having a ton more health kits when you're in areas with lots of spider splicers - very handy.

Now that you have the camera, never start shooting enemies right away. Take as many pictures as possible until your score stops going up fairly well each time. You get damage bonuses as well as special plasmids for taking pictures. There are 5 research levels for each enemy(including sisters and big daddies - don't forget to take pictures of them before fights!!) - dmg up - plasmid - dmg up - plasmid - dmg up <---those are what you get for each research level per enemy(at least, if my memory serves me well). Seriously, though, abuse the hell out of that camera and watch your film 'ammo'.

I went through very slowly the first time as well, don't worry about it. I always hated using the vitachambers, so I wound up reloading saves a lot and there were some fights where I got stuck and had to get imaginative in how to kill things. Once you get into the late game, don't take too long to explore everything, or you'll start to get bored. Bioshocks only weakness (imo) is that in late game you're tired of the same enemies just getting buffed up and you are pretty over-powered and well-funded - so you never run out of ammo as long as you find vending machines or u-invent stands.

I need another runthrough with this game. I didn't use the camera nearly enough and so missedo ut on some plasmids. I also abused the vitachambers, which is effective but feels suspiciously like chaeting.

I didn't know you could research Little Sisters until like the end of the game.

Stupid memo said "Research 'enemies'".

Yeah I only got to midstage with Wrench only... I need to finish this game before BioShock 2 comes out.

Also on BioShock 2, anyone think they're just making things up as they go along?

Like I just found out now there were multiple Big Sisters, rather than the solitary one which is Sofia Lamb... Who now isn't a Big Sister but the new Andrew Ryan/Atlas/Fountaine.

best use I found for rocket launcher turrets was to hack them and lure big daddies towards them.

I found a glitch sister finally, But I didn't have enough supplies to kill the daddy. I went back to the vending machine and when I returned, no sister. I guess that means I fail. Oh well.

on the metro level now...still loving every second of this game. I got to the point where I'm in the research lady's office and need to fight my way back out past a pile of security stuff. Bleugh.

Cool stuff lately:

got the chemical thrower and grenade launcher, both of which are great by the way. 3 proximity mines and an armor-piercing clip from the damage-increased machine gun and a rosie is basically toast now. I killed my first daddy without dying the other night.

Dealt with my first round of houdinis, but considering they last about as long as it takes me to unload 2 shells from my damage-increased shotgun, they aren't a big hassle anymore. First guy was tough though. That research camera makes a humungous difference when dealing with the assorted enemies. Once you get a couple of damage-ups they really stop being an issue. I already almost maxed research on the spider splicers and the regular ledhead / thuggish splicers. About halfway through the houdinis and still working on the daddies. What else can you research enemy-wise? Sisters? turrets?

Researching sisters gives you an increase in max health iirc. Researching Houdinis is the most important for the easy-mode chameleon tonic.
